Cervical leukoplakia in Ghana: Unveiling diagnostic challenges and informing low-resource screening strategies.

A retrospective study of 13,861 women screened for cervical cancer in Ghana between 2016 and 2024 found that cervical leukoplakia occurred in 0.9% of women, and critically, 93.3% of these cases showed no acetowhitening on Visual Inspection with Acetic Acid (VIA) or mobile colposcopy, meaning the most widely used low-resource screening method would routinely fail to flag them. Among the 21 women (17.5%) who underwent LEEP excision, histopathology confirmed high-grade disease in 3 cases (CIN III), demonstrating that leukoplakia carries real oncological risk even in the absence of the classic acetowhitening sign. High-risk HPV positivity was the strongest independent risk factor for leukoplakia (adjusted OR: 30.27; 95% CI: 3.56–257.37), followed by HIV infection (adjusted OR: 8.27; 95% CI: 1.18–57.69), findings that call for integrated point-of-care triage tools and simplified management protocols tailored to low-resource settings.

Premature mortality and years of potential productive life lost due to HPV-attributable cancers in Brazil and Mexico: 2011 Versus 2023.

A population-based study of HPV-attributable cancer mortality in Brazil and Mexico found a persistent and growing burden of premature death between 2011 and 2023, with total HPV-attributable deaths rising from 6,515 to 9,533 in Brazil (years of life lost +44.6%) and from 4,303 to 4,997 in Mexico (years of life lost +21.4%). Penile cancer years of life lost increased by 54.8% in Brazil and 37.7% in Mexico over this period, while anal cancer showed the largest relative increases — up to +203.8% among Brazilian men — and oropharyngeal cancer years of life lost rose 33.5% in Brazil and 74.6% in Mexico. By 2023, years of potential productive life lost totaled 97,404 years in Brazil and 51,735 years in Mexico, representing increases of 43.9% and 26.7% relative to 2011, respectively. The authors conclude that expanded HPV vaccination, systematic screening, timely diagnosis, and equitable treatment access are urgently needed across all HPV-associated cancer sites in both countries.

Cytotoxic phenanthrene and racemosol derivatives from Bauhinia malabarica roots: network pharmacology and in silico mechanistic insights.

Racemosol, a natural compound isolated from the roots of the medicinal plant Bauhinia malabarica, demonstrated potent cytotoxic activity against HeLa cervical cancer cells with an IC50 of 4.51 μM and induced cell death via apoptosis in a dose-dependent manner. Researchers isolated ten compounds in total — including three previously unreported phenanthrene and racemosol derivatives named malathrene A, malasol A, and bauhinol B — and used network pharmacology together with molecular docking to investigate the mechanism of action. The computational analyses revealed that racemosol likely modulates the PI3K-Akt and MAPK signaling pathways and interacts with the ATP-binding site of the EGFR kinase domain, suggesting its potential as a tyrosine kinase inhibitor. These findings highlight racemosol as a promising plant-derived lead compound warranting further pharmacological development for cervical cancer therapy.
