Kinase inhibitors (TKIs)
Kinase inhibitors are small-molecule drugs, usually taken as tablets, that block enzymes (kinases) relaying growth signals inside cancer cells. When a tumor depends on an overactive kinase — such as BCR-ABL in chronic myeloid leukemia or KIT in gastrointestinal stromal tumors — blocking it can slow or stop the growth of cancer cells. Eligibility depends on finding the specific abnormality in the tumor, and cells can develop resistance over time.

Used in
tyrosine kinase inhibitors in Philadelphia-positive ALL; FLT3 inhibitors in selected AML
BRAF and MEK inhibitors are standard for tumours with a BRAF V600 mutation
option in advanced disease refractory to radioiodine or with RET/BRAF alterations
anti-angiogenic kinase inhibitors are established in advanced disease
sorafenib and lenvatinib are established options in advanced disease
standard for tumours with EGFR, ALK or ROS1 alterations, guided by molecular testing
CDK4/6 inhibitors with hormone therapy are standard in advanced hormone receptor-positive disease
