Tissue-Free Circulating Tumor DNA Assay and Patient Outcome in a Phase III Trial of FOLFOX-Based Adjuvant Chemotherapy (Alliance N0147)
A study published in the Journal of Clinical Oncology evaluated a tissue-free circulating tumor DNA (ctDNA) assay for predicting patient outcomes in the context of FOLFOX-based adjuvant chemotherapy for colon cancer, using data from the large phase III Alliance N0147 trial. The tissue-free approach is notable because it does not require prior tumor sequencing to design a personalized ctDNA panel, potentially making liquid biopsy monitoring more accessible and scalable. Results from this validation in a randomized phase III cohort provide high-level evidence on the prognostic or predictive value of ctDNA in guiding post-surgical treatment decisions. This work strengthens the case for integrating ctDNA-based minimal residual disease detection into routine clinical practice for colorectal cancer patients receiving adjuvant therapy.
