Bridging the Gap: Precision Oncology for Cancers of Unknown Primary

A new review in the Journal of Clinical Oncology examines how precision oncology approaches — including molecular profiling and genomic classifiers — can improve diagnosis and treatment of cancers of unknown primary (CUP). CUP accounts for 3–5% of all malignancies and historically carries poor prognosis due to empiric, non-targeted therapy. The article discusses how tissue-of-origin assays and next-generation sequencing may enable site-specific treatment strategies, potentially bridging the gap between CUP and identified primary tumors.
