Teclistamab versus lenalidomide-dexamethasone in high-risk smoldering multiple myeloma: a randomized phase 2 trial

In the randomized phase 2 ImmunoPRISM trial, teclistamab achieved higher rates of complete clinical responses than lenalidomide–dexamethasone in patients with high-risk smoldering multiple myeloma. This is a notable signal for early intervention with a bispecific T-cell engager (BCMAxCD3) in a pre-malignant plasma cell disorder. However, longer follow-up is needed to determine whether these deeper responses translate into durable prevention of progression to active multiple myeloma.
