Magrolimab Plus Azacitidine Versus Placebo Plus Azacitidine in Patients With Untreated Higher-Risk Myelodysplastic Syndromes: The Phase III ENHANCE Study

The phase 3 ENHANCE trial met neither of its dual primary endpoints: adding the anti-CD47 antibody magrolimab to azacitidine did not improve outcomes in 539 treatment-naive patients with intermediate- to very-high-risk myelodysplastic syndromes. The complete remission rate was 21.3% with magrolimab plus azacitidine versus 23.6% with placebo plus azacitidine, and median overall survival was 15.9 versus 18.6 months. Grade 3 or higher adverse events, serious and fatal adverse events, and discontinuations due to adverse events were all more frequent in the magrolimab arm.
