Ifebemtinib plus garsorasib in previously treated metastatic colorectal cancer with KRASG12C mutation: a multicentre, randomised, phase 1b/2 trial

Adding ifebemtinib to garsorasib did NOT significantly improve response rate in a very small study: the randomised portion included just 36 patients with previously treated KRAS G12C-mutated metastatic colorectal cancer. The confirmed objective response rate was 38.9% (95% CI 17.3-64.3) versus 16.7% (95% CI 3.6-41.4) with garsorasib alone, but the 22.2-percentage-point difference had a confidence interval spanning zero (95% CI -7.7 to 49.1; one-sided p=0.068). Grade 3 adverse events occurred in 33% versus 28% of patients, with no treatment-related deaths. The authors treat the result as grounds for further study rather than evidence that the combination works.
